Start by verifying three baseline credentials: GMP certification from a recognized authority (NMPA, FDA, or EU), active DMF or CEP filings for the API you need, and export history to regulated markets. Beyond documents, evaluate the supplier's communication responsiveness, willingness to share batch CoAs before an order, third-party audit history, and whether they participate in industry consortiums like Rx-360. Comparing 2-3 manufacturers on these dimensions before committing to a purchase order substantially reduces sourcing risk.
At minimum, request: (1) a current Certificate of Analysis (CoA) from a recent production batch, (2) a Method of Analysis document detailing the test procedures, (3) evidence of GMP compliance (GMP certificate or inspection report), (4) the manufacturer's DMF status and CEP status if applicable, (5) a Product Quality Standard Sheet or Specification Sheet, and (6) an MSDS. For regulated markets, request documentation on the manufacturer's regulatory filing history with the FDA or EDQM. A supplier who provides these documents without hesitation is usually more reliable.
MOQs vary by product type and supplier tier. For high-value APIs like UDCA, many suppliers accommodate 1-5 kg trial orders for initial qualification, while commercial-scale batches typically start at 25 kg. For commodity APIs such as paracetamol or metformin, MOQs are often 100 kg or more. Veterinary APIs like spectinomycin HCl typically have MOQs of 25 kg (one drum). Suppliers targeting regulated markets generally offer more flexibility on trial quantities than pure commodity exporters. Note that unit prices decrease considerably as order volumes increase.

A supplier's willingness to support third-party testing is itself a strong reliability signal.
Both models have merit. Direct manufacturer sourcing can offer lower pricing and direct access to technical staff, but typically involves higher minimum orders and limited regulatory documentation support. Quality & Certification

The Quality System is mandatory in every ICH Q7 inspection and is the foundation for all other systems. Focus on the independence of the Quality Unit, annual Product Quality Reviews covering impurity trends, formal change control for process or supplier changes, and a functioning deviation/CAPA system. A weak Quality System undermines everything else — a facility with good equipment but no quality oversight will eventually ship bad product.
Based on FDA warning letter trends and APIC guidance, the most frequent findings are: (1) inadequate cleaning validation — especially cross-contamination risk in shared equipment; (2) poorly justified API Starting Material definition without scientific rationale per ICH Q11; (3) data integrity deficiencies in HPLC systems — shared logins, disabled audit trails, and unreviewed manual integrations; (4) incomplete OOS investigations — closing failures as "laboratory error" without proper root cause analysis. Third-party audit reports from accredited organizations like Eurofins, SGS, or Intertek are useful for initial screening and can reduce the scope of your own audit. However, most regulators expect the Marketing Authorization Holder to conduct or commission a direct audit of their API suppliers. A third-party report alone may not satisfy your GMP obligations. A DMF (Drug Master File) is a confidential document filed with the FDA containing detailed manufacturing, processing, and quality information about an API. Products & Applications

UDCA (ursodeoxycholic acid) is the unconjugated free acid. TUDCA (tauroursodeoxycholic acid) is the taurine conjugate of UDCA. In the body, UDCA is conjugated with taurine (predominantly) or glycine in the liver before secretion into bile. TUDCA is more water-soluble than UDCA and is sometimes used in research settings and as a dietary supplement. Pharmaceutical products for human therapeutic use are formulated with UDCA, not TUDCA, and UDCA capsules/tablets are the registered drug product form in all major pharmacopoeias.
TUDCA (tauroursodeoxycholic acid) is synthesized from UDCA through conjugation with taurine. UDCA is extracted and purified from bovine bile, then chemically conjugated with taurine to form TUDCA. The manufacturing process involves four stages: (1) extraction of cholic acid from bovine bile, (2) chemical synthesis through epimerization to UDCA, (3) conjugation with taurine via an amide bond formation reaction, and (4) purification to pharmaceutical or supplement grade (typically over 98% purity by HPLC). TUDCA is primarily used for liver health support and has been studied for multiple therapeutic applications: (1) hepatoprotection in cholestatic liver diseases, where it displaces toxic hydrophobic bile acids and stimulates bile flow; (2) reduction of liver enzymes (ALT, AST) in chronic hepatitis and metabolic liver conditions; (3) neuroprotection in preclinical models of Alzheimer's disease, Parkinson's disease, and ALS, based on its ability to cross the blood-brain barrier and reduce endoplasmic reticulum stress; (4) metabolic benefits including improved insulin sensitivity as demonstrated in proof-of-concept human studies.
